Paper 13324-25 NADH, FAD and TRP metabolic FLIM: tuneable excitation from 400 nm – 1300 nm for 1p vs. 2p imaging of the same sample area (Invited Paper) 27 January 2025 • 10:45 AM - 11:00 AM PST | Moscone South, Room 301 (Level 3) FLIM experiments typically rely on basic assumptions for interpretation. First, the absorption and emission spectra may not translate from pure samples to ‘real’ biological samples. Second, a chosen excitation may excite a plethora of fluorophores other than the target. It is, therefore, crucial for comparative studies to have precise tuneable control of the excitation wavelength. Third, the absorption spectra for 1-p and 2-p excitation of most fluorophores are not exactly a factor 2 apart, a common assumption. We present a tunable (400 nm – 1300 nm) femtosecond FLIM microscopy system to compare 1-p and 2-p results from the same sample area. Presenter Julius Heitz Becker & Hickl GmbH (Germany)

Paper 13324-15 Metabolic NADH FLIM: precise, turnkey quantitative FLIM, FLIM-FRET and more in zen for the Zeiss LSM980 (Invited Paper) 26 January 2025 • 3:50 PM - 4:10 PM PST | Moscone South, Room 301 (Level 3) We present NADH metabolic imaging FLIM results and an intuitive turnkey FLIM solution for the Zeiss LSM980 – controlled in the Zen software. Based on the full precision and efficiency of Becker&Hickl GmbH TCSPC technology, data is recorded, sent through the SPCimage NG analysis pipeline, and results are returned to Zen for comparison. Advantages include: direct visualisation of and access to all fit parameters, lifetime analysis and multi-parameter segmentation of 2D/3D data sets, and unsurpassed lifetime resolution with a system IRF down to ~20ps @203 fs bins to resolve even the smallest lifetime differences. 325 and 340 nm UV LEDs for Biological Applications
Our family of 325 and 340 nm UV LEDs and arrays from Violumas are made for biological applications such as biological analysis, spectroscopy, and sensing. Producing wavelengths of 325 nm and 340 nm, respectively, the LEDs come in 2×2 modules in a chip-on-board format and have a LT70 of greater than 6000 hours. With four chips encapsulated under the same lens with a specially developed index boosting solution, the modules feature a 10.2% wall-plug efficiency at 340 nm and 3% wall-plug efficiency at 325 nm.
